Naloxone fluorescein acetate is a fluorescently labeled naloxone probe used for detecting Opioid Receptor expression. Naloxone fluorescein acetate generates fluorescent signals via flow cytometry, enabling indirect evaluation of classical opioid receptor expression. Naloxone fluorescein acetate is applicable to studies related to opioid receptor expression in canine tumor cells.

Protocol Guide (The following is our recommended protocol. This protocol is for reference only and should be modified according to your specific needs).
1. Stock Solution Preparation
1.1 Solvent: Naloxone fluorescein acetate is dissolved in DMSO.
1.2 Recommended concentration: A high-concentration stock solution of 1-10 mM is generally recommended.
2. Working Solution Preparation
2.1 Diluent: Dilute the dye with PBS/1% FBS for cell resuspension.
2.2 Working concentration: The concentration actually used in the literature is 0.05 nM.
3. Staining Procedure
3.1 Sample types: Canine transitional cell carcinoma K9TCC, canine osteosarcoma Abrams, and canine hemangiosarcoma DAL-4 cells.
3.2 Incubation conditions: Cells are incubated with Naloxone fluorescein acetate (0.05 nM) at room temperature in the dark for 30 min.
3.3 Washing steps: After incubation, cells are washed, centrifuged, and resuspended in ice-cold PBS/1% FBS.
4. Control Setup
4.1 Control group: Cells pretreated with normal saline serve as the control to evaluate the baseline proportion of opioid receptor-positive cells.
4.2 Experimental treatment group: Cells pretreated with Doxorubicin (HY-15142A) are used to compare changes in the proportion of opioid receptor-positive cells; K9TCC, Abrams, and DAL-4 cells are pretreated with 0.5, 0.15, and 0.05 μg/mL Doxorubicin for 72 h, respectively.
5. Detection and Analysis
5.1 Instrument: A flow cytometer is used for analysis.
5.2 Result analysis: The proportion of fluorescence-positive cells is analyzed to indirectly evaluate opioid receptor expression. Approximately 35%-80% of control cells are receptor-positive; after Doxorubicin pretreatment, the receptor-positive proportion of K9TCC and Abrams cells increases nearly 2-fold and rises above 90%, while DAL-4 cells show no significant increase.
