PA Janelia Fluor 549, SE (PA-JF549-NHS) is a bright photoactivatable fluorophore of JF549,SE (JF549,NHS). JF549,SE (JF549,NHS) is a fluorescent dye with the absorption maximum (λab (max)) of 549 nm and emission maximum (λem (max)) of 571 nm. Janelia Fluor fluorescent dyes can be chemically conjugated to the specific HaloTag substrate (a chloroalkane ligand) to form a fluorescent ligand, rather than binding directly to the HaloTag protein itself.

Biological Activity
Janelia Fluor fluorescent dyes do not inherently possess HaloTag-binding activity; however, reactive groups such as NHS esters serve as reaction sites for covalent coupling-via chemical synthesis-to an amino-functionalized HaloTag chloroalkane ligand scaffold, thereby yielding a fluorescent HaloTag ligand.
PA Janelia Fluor is the integration of a four-member azetidine ring, which reduces the photobleaching and photodamage, improves the brightness and photostability of the fluorescent dye. PA Janelia Fluor exhibits cell membrane permeability that can be used for live cell imaging[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
