Description
The ADAM17/TACE protein cleaves TNF-α, releasing it from the membrane and activating various cell surface proteins such as JAM3, ACE2, and the p75 TNF receptor. It also triggers the Notch pathway by cleaving Notch into membrane-associated fragments. ADAM17/TACE Protein, Mouse (HEK293, His) is the recombinant mouse-derived ADAM17/TACE protein, expressed by HEK293 , with C-His labeled tag.
Background
ADAM17/TACE Protein is responsible for cleaving the membrane-bound precursor of TNF-alpha, converting it into its mature soluble form. It also plays a crucial role in the proteolytic release of various cell-surface proteins, including JAM3, ACE2, p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1, and the amyloid precursor protein. Additionally, ADAM17/TACE activates the Notch pathway by mediating the cleavage of Notch, generating a membrane-associated intermediate fragment known as notch extracellular truncation (NEXT). In hemostasis, it contributes by shedding the platelet glycoprotein Ib alpha chain (GP1BA), while also facilitating the proteolytic cleavage and release of secreted forms of LAG3 and IL6R. Furthermore, ADAM17/TACE mediates the proteolytic cleavage and shedding of FCGR3A upon stimulation of natural killer cells, allowing for enhanced NK cell motility and detachment from opsonized target cells.
Verified Bioactivity
Measured by its ability to cleave 10μM fluorogenic peptide substrate Mca-PLAQAV-Dpa-RSSSR-NH2 that incubate at room temperature in kinetic mode for 5 minutes. The specific activity is 1818.15 pmol/min/µg.
