Description
ALK Protein, Human (L1196M, Biotinylated, sf9, Avi) is the recombinant human-derived ALK, expressed by Sf9 insect cells, with Avi labeled tag.
Background
ALK is a neuronal receptor tyrosine kinase transiently expressed in specific nervous system regions, playing crucial roles in neurogenesis and differentiation. It also regulates energy homeostasis by controlling white adipose tissue lipolysis and sympathetic tone (By similarity). Activated by ALKAL2 (but not ALKAL1), it initiates intracellular signaling through MAPK pathway activation, phosphorylating targets like CBL, FRS2, IRS1, and SHC1. ALK may also be modulated by pleiotrophin (PTN) and midkine (MDK), which promote anti-apoptotic signaling and cell proliferation via MAPK/PI3K pathways. Additionally, ALK drives NF-κB activation through IRS1-AKT, essential for MDK-mediated autocrine growth and survival.
Verified Bioactivity
The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecencelabeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 0.39 nM, the conversion rate is about 40%.
