Biological Activity

Description

The B7-H4 protein acts as a negative regulator, inhibiting T cell-mediated immune responses, including activation, proliferation, cytokine production, and cytotoxicity. When expressed on tumor macrophages, it cooperates with regulatory T cells to suppress tumor-associated antigen-specific T cell immunity. B7-H4 Protein, Human (Biotinylated, HEK293, His-Avi) is the recombinant human-derived B7-H4 protein, expressed by HEK293 , with C-Avi, C-His labeled tag.

Background

B7-H4 protein functions as a negative regulator of the T-cell-mediated immune response, exerting inhibitory effects on T-cell activation, proliferation, cytokine production, and the development of cytotoxicity. Particularly significant when expressed on the cell surface of tumor macrophages, B7-H4, in collaboration with regulatory T-cells (Treg), plays a crucial role in suppressing tumor-associated antigen-specific T-cell immunity. Additionally, B7-H4 is implicated in the promotion of epithelial cell transformation, indicating its multifaceted involvement in modulating immune responses and cellular processes associated with tumor development.

Verified Bioactivity

1. Measured by its binding ability in a functional ELISA. When immobilized Anti-B7-H4 Antibody at 1μg/ml (100μl/well), can bind Biotinylated Human B7-H4, His Tag and the EC50 is < 65 ng/mL.
2. Loaded Puxitatug (HY-P990059) on AHC2 biosensor, can bind B7-H4 Protein, Human (Biotinylated, HEK293, His-Avi) with an affinity constant of 4.217E-09 M as determined in BLI assay.
3. Loaded Emiltatug (HY-P990918) on AHC2 biosensor, can bind B7-H4 Protein, Human (Biotinylated, HEK293, His-Avi) with an affinity constant of 8.115E-09 M as determined in BLI assay.
