Background
EAAT3 is a sodium-dependent, high-affinity amino acid transporter that mediates the uptake of L-glutamate, L-aspartate, and D-aspartate, with additional capability to transport L-cysteine. It functions as a symporter, co-transporting one amino acid molecule with two or three Na(+) ions and one proton while counter-transporting one K(+) ion. The protein also facilitates Cl(-) flux uncoupled from amino acid transport to prevent negative charge accumulation during aspartate/Na(+) symport. It plays a key role in renal reabsorption of L-glutamate and L-aspartate. By similar functions: it contributes redundantly to synaptic glutamate clearance for neurotransmission termination, participates in glutathione biosynthesis and oxidative stress protection via L-glutamate/L-cysteine transport, and is negatively regulated by ARL6IP5.
