ITK Protein, Human (Active, 269a.a, sf9, His, GST)

Cat. No.: HY-P705930
Handling Instructions Technical Support

ITK protein is an important tyrosine kinase that regulates adaptive immune responses by affecting the development and function of T cells and NKT cells. Upon T cell receptor (TCR) activation, ITK undergoes LCK-mediated phosphorylation, leading to its activation. ITK Protein, Human (Active, 269a.a, sf9, His, GST) is the recombinant human-derived ITK, expressed by Sf9 insect cells, with His and GST labeled tag.

Description

ITK protein is an important tyrosine kinase that regulates adaptive immune responses by affecting the development and function of T cells and NKT cells. Upon T cell receptor (TCR) activation, ITK undergoes LCK-mediated phosphorylation, leading to its activation. ITK Protein, Human (Active, 269a.a, sf9, His, GST) is the recombinant human-derived ITK, expressed by Sf9 insect cells, with His and GST labeled tag.

Background

ITK protein, a crucial tyrosine kinase, plays an indispensable role in the regulation of the adaptive immune response by orchestrating the development, function, and differentiation of conventional T-cells and nonconventional NKT-cells. Upon activation of the T-cell receptor (TCR) by antigen-presenting cells (APC), a phosphorylation cascade ensues, leading to the recruitment of ITK to the cell membrane in close proximity to the stimulated TCR receptor. Subsequently, ITK undergoes phosphorylation by LCK, resulting in its autophosphorylation and full activation. Once activated, ITK phosphorylates PLCG1, activating this lipase and initiating the cleavage of its substrates. This process triggers the release of calcium from the endoplasmic reticulum into the cytoplasm, facilitating the translocation of the nuclear activator of activated T-cells (NFAT) into the nucleus for transcriptional regulation. Additionally, ITK phosphorylates essential adapter proteins like the linker for activation of T-cells/LAT protein and LCP2, recruiting a myriad of signaling molecules, including VAV1, ultimately leading to lymphokine production, T-cell proliferation, and differentiation. ITK is crucial for TCR-mediated calcium response in gamma-delta T-cells and may play a role in modulating the transcriptomic signature in the Vgamma2-positive subset of immature gamma-delta T-cells. Furthermore, ITK phosphorylates TBX21 at 'Tyr-530,' mediating its interaction with GATA3. This multifaceted involvement underscores the pivotal role of ITK in shaping diverse facets of the adaptive immune response.

Biological Activity

The activity was measured by off-chip mobility shift assay(MSA). The enzyme was incubated with fluorecencelabeled substrate and Mg(or Mn)/ATP. The phosphorylated and unphosphorylated substrates were separated and detected by MSA device. When the protein concentration is 6.25 nM, the conversion rate is about 20%.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
