NRG1-beta 1 Protein, Human (246a.a, His)
Based on 1 Customer Validation
NRG1 is a direct ligand of the ERBB3 and ERBB4 tyrosine kinase receptors. NRG1 can induce the phosphorylation and activation of MAPK3/ERK1, MAPK1/ERK2 and AKT1. NRG1 can induce cell growth, differentiation and proliferation. NRG1-beta 1 Protein, Human (246a.a, His) is the recombinant human-derived NRG1-beta 1 protein, expressed by E. coli , with N-6*His labeled tag.

Background
Pro-neuregulin-1, membrane-bound isoform (NRG1) is a direct ligand of the ERBB3 and ERBB4 tyrosine kinase receptors. Several isomers of NRG1 can induce the growth and differentiation of epithelial cells, glial cells, neuronal cells and skeletal muscle cells. The induced expression of acetylcholine receptors in synaptic vesicles during the formation of neuromuscular junction stimulates the germination and lactation of breast lobular alveoli and induces the differentiation of breast tumor cells. Stimulate Schwann cell proliferation; It affects the development of the heart muscle. NRG1 is also a ligand for integrins and binds (via the EGF domain) the integrins ITGAV: ITGB3 or ITGA6:ITGB4. NRG1 can induce the phosphorylation and activation of MAPK3/ERK1, MAPK1/ERK2 and AKT1[1][2][3][4].
Verified Bioactivity
Measured in a serum-free cell proliferation assay using MCF-7 human breast cancer cells. The ED50 for this effect is 33.21 ng/mL, corresponding to a specific activity is 3.01×104 units/mg

Product Properties
Lyophilized powder
Lyophilized from a 0.22 μm filtered solution of 4 mM HCl or 50 mM Tris-HCl, 300 mM NaCl, 200 mM arginine, pH 8.0.
Note: For SPR assay, please replace the buffer. Primary amine components (e.g., Tris, imidazole) can affect protein-coupled chips.
<1 EU/μg, determined by LAL method.
It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).
Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
Room temperature in continental US; may vary elsewhere.
