Biological Activity
PGAM5 Protein, Human (His) is the recombinant human-derived PGAM5, expressed by E. coli , with C-6*His labeled tag. ,
PGAM5 is a mitochondrial serine/threonine phosphatase that dephosphorylates various substrates, playing roles in processes like cellular senescence and mitophagy. It modulates mitochondrial dynamics by dephosphorylating DNM1L/DRP1 to promote fission and stress-sensitively dephosphorylating MFN2 to protect it from ubiquitination, aiding mitochondrial network formation. Independently of PARKIN, it regulates mitophagy by interacting with and dephosphorylating FUNDC1 (which binds LC3). PGAM5 also forms a tertiary complex with KEAP1 and NRF2 to regulate anti-oxidative responses, and acts as a RIPK3 target to recruit the RIPK1-RIPK3-MLKL necrosome to mitochondria, controlling necroptosis.
Human PGAM5 immobilized on CM5 Chip can bind LFHP-1c with an affinity constant of 425.8 nM as determined in a SPR assay.
