Background
PH20/SPAM1 protein plays a crucial role in sperm-egg adhesion during fertilization. In the intricate process of fertilization, sperm encounters a layer of cumulus cells surrounding the egg before reaching the zona pellucida. These cumulus cells are embedded in a matrix rich in hyaluronic acid, formed in preparation for ovulation. PH20/SPAM1 facilitates sperm penetration through the cumulus cell layer by digesting hyaluronic acid. By engaging in this enzymatic activity, PH20/SPAM1 contributes to the essential steps of fertilization, enabling the sperm to navigate through the cumulus cell barrier and reach the zona pellucida for successful fertilization to occur.

The activity of Human Hyaluronidase PH-20 Protein, His Tag is measured by its ability to hydrolyze HA in turbidimetric assay (45 minute assay). The specific activity is >60,000 U/mg. (Unit Definition: One unit of Hyaluronidase activity will cause a change in A600 of 0.330 per minute at pH 5.35 at 37°C in a 2.0 mL reaction mixture)
