Background
The PICALM gene encodes a clathrin assembly protein crucial for recruiting clathrin and adaptor protein complex 2 (AP2) to cell membranes during coated-pit formation and clathrin-vesicle assembly. Its role may extend to regulating the size of the clathrin cage, thus determining the amount of membrane to be recycled. This protein is implicated in AP2-dependent clathrin-mediated endocytosis, particularly at the neuromuscular junction. Notably, chromosomal translocation involving this gene is associated with various hematological malignancies. Additionally, polymorphisms in this gene are linked to Alzheimer's disease risk. The gene exhibits ubiquitous expression, with notable levels observed in fat, appendix, and 25 other tissues. Multiple alternatively spliced transcript variants encoding different isoforms have been identified.
