Background
PICK1 is a probable adapter protein that binds to and organizes the subcellular localization of various membrane proteins containing PDZ recognition sequences. It is involved in the clustering of multiple receptors, potentially by acting at the receptor internalization level. PICK1 plays a role in synaptic plasticity by regulating the trafficking and internalization of AMPA receptors and may be regulated upon PRKCA activation. Additionally, PICK1 may modulate ASIC1/ASIC3 channels and regulates actin polymerization by inhibiting the actin-nucleating activity of the Arp2/3 complex, a function competitive with nucleation promoting factors and linked to neuronal morphology regulation and AMPA receptor (AMPAR) endocytosis. Through interaction with the Arp2/3 complex, PICK1 is involved in regulating synaptic plasticity at excitatory synapses and is required for spine shrinkage during long-term depression (LTD). PICK1 also regulates astrocyte morphology, antagonizing the function of the Arp2/3 complex activator WASL/N-WASP.
