Plexin B2 (PLXNB2) serves as a cell surface receptor for SEMA4C, SEMA4D, and SEMA4G, playing a crucial role in cell-cell signaling. It is involved in glutamatergic synapse development and is required for SEMA4A-mediated excitatory synapse development. Binding to class 4 semaphorins induces downstream activation of RHOA and phosphorylation of ERBB2 at 'Tyr-1248.' PLXNB2 is essential for normal differentiation and migration of neuronal cells during brain corticogenesis, contributing to normal embryonic brain development, and regulating the migration of cerebellar granule cells in the developing brain. Additionally, it plays a pivotal role in RHOA activation, leading to changes in the actin cytoskeleton, and participates in axon guidance, invasive growth, and cell migration. PLXNB2 may modulate the activity of RAC1 and CDC42, existing as both a monomer and a heterodimer with PLXNB1. Furthermore, it interacts with SEMA4C, SEMA4D, SEMA4G, MET, ARHGEF11, ARHGEF12, and possibly MST1R.
