Background
ROCK2 is a protein kinase that serves as a key regulator of the actin cytoskeleton and cell polarity. It modulates smooth muscle contraction, actin cytoskeleton organization, stress fiber and focal adhesion formation, neurite retraction, cell adhesion, and motility by phosphorylating proteins such as ADD1, BRCA2, CNN1, EZR, DPYSL2, EP300, MSN, MYL9/MLC2, NPM1, RDX, PPP1R12A, and VIM. Additionally, ROCK2 phosphorylates SORL1 and IRF4 and acts as a negative regulator of VEGF-induced angiogenic endothelial cell activation. During myogenic differentiation, it positively regulates the activation of p42/MAPK1-p44/MAPK3 and p90RSK/RPS6KA1. ROCK2 plays a crucial role in the timely initiation of centrosome duplication and inhibits keratinocyte terminal differentiation. It may regulate eyelid closure and ventral body wall formation by organizing actomyosin bundles. In the hippocampus, ROCK2 is critical for regulating spine and synaptic properties and contributes to the circadian rhythm of aortic myofilament Ca2+ sensitivity and vascular contractility by modulating myosin light chain phosphorylation.
