TIGIT Protein, Human (HEK293, Fc)

TIGIT Protein, Human (HEK293, Fc) produced in HEK293 cells with the C-termimal human IgG1 Fc fragment. T-cell immunoreceptor with Ig and ITIM domains (TIGIT) is also known as V-set and immunoglobulin domain-containing protein 9 (VSIG9), V-set and transmembrane domain-containing protein 3 (VSTM3). It belongs to single-pass type I membrane protein containing an immunoglobulin variable domain, a transmembrane domain and an immunoreceptor tyrosine-based inhibitory motif (ITIM).

T cell Ig and ITIM domain (TIGIT) is a transmembrane glycoprotein belonging to a poliovirus receptor family of type 1 proteins that also binds to CD155 and CD112 ligands. TIGIT is expressed on peripheral memory and regulatory CD4+ T cells and NK cells and is upregulated following activation on naive CD4+ T cells. CD155 is a high-affinity receptor for TIGIT expressed on monocytes and CD11c+ human dendritic cells (DCs). TIGIT contains an Ig-like V-type domain and an ITIM in its cytoplasmic domain, suggesting that receptor occupancy may trigger a negative signaling event^[1].

5 µg/mL (100 µL/well) of immoblized recombinant human CD155-Fc can bind human Biotin-TIGIT-Fc with a linear range of 6.1-48.83 ng/mL.

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
