Background
UBE2C, a pivotal component of the ubiquitin-proteasome system, functions as an E2 ubiquitin-conjugating enzyme, playing a central role in the covalent attachment of ubiquitin to target proteins. In vitro, UBE2C demonstrates catalytic proficiency in facilitating 'Lys-11'- and 'Lys-48'-linked polyubiquitination, showcasing its versatility in ubiquitin chain formation. Crucially, UBE2C acts as an indispensable factor in the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated ubiquitin ligase pivotal for governing mitotic progression. Its role in initiating 'Lys-11'-linked polyubiquitin chains on APC/C substrates underscores its significance in orchestrating the degradation of specific proteins by the proteasome, thereby promoting a timely and controlled exit from mitosis.
Verified Bioactivity
The ubiquitin conjugating activity of UBE2C was validated through its ability to catalyse the generation of polyubiquitin chains in the presence of the E1 activating enzyme UBE1 and Bodipyubiquitin. Incubation of UBE2C for 60 minutes at 37˚C in the presence of Bodipy-ubiquitin, UBE1 and ATP was compared alongside two control reactions with either ATP or UBE2C excluded from the reaction. Ubiquitin conjugates were identified by the migration of the Bodipy-ubiquitin band and these were observed only in the presence of ATP and UBE2C.
