(S)-Benzyl pyrrolidine-2-carboxylate hydrochloride is a key raw material for the synthesis of apoptosis inhibitor protein (IAP) antagonists and SMAC mimic tripeptide fragments (Boc-MeAla-Tle-Pro-OH). (S)-Benzyl pyrrolidine-2-carboxylate hydrochloride can be used to synthesize divalent quinazoline IAP antagonists.

In Vitro
(S)-Benzyl pyrrolidine-2-carboxylate hydrochloride (compound 14) can undergo an amide coupling reaction with Boc-Tle-OH in the HATU/DIPEA/DCM system to generate Boc-Tle-Pro-OBn. After de-Boc by 4M HCl/dioxane, it is coupled with Boc-MeAla-OH and finally debenzylated by H2/Pd-C to obtain tripeptide fragment 16, which is used for the synthesis of divalent quinazoline IAP antagonists[1].
