Biological Activity
Suc-Ala-Glu-Pro-Phe-pNA functions as a substrate for recombinant human Pin1 in a cell-free protease-coupled PPIase assay, enabling measurement of Pin1 isomerase activity via pNA release[1].
Suc-Ala-Glu-Pro-Phe-pNA (Suc-AEPF-pNA) (600 nM) specifically elutes phage bound to the Pin1 PPIase domain active site, demonstrating it interacts with the domain's active site[2].
Suc-Ala-Glu-Pro-Phe-pNA (Suc-AEPF-pNA) (0.33 ± 0.06 mM) serves as a substrate for GST-tagged Pin1 PPIase activity, with a KM of 0.33 mM and a kCAT of 7.3 s?1[2].
Suc-Ala-Glu-Pro-Phe-pNA (2.5 mM) acts as a substrate for recombinant human Pin1's PPIase activity, which is progressively inhibited as Pin1 is phosphorylated by PKA over time, with ~85% inhibition of Pin1's ability to isomerize Suc-Ala-Glu-Pro-Phe-pNA observed after 1 h of PKA pre-incubation[3].
