Sulfo-Cy5-TCO is a water-soluble sulfonated Cy5 fluorescent reagent containing trans-cyclooctene (TCO). Sulfo-Cy5-TCO undergoes an inverse electron demand Diels‑Alder reaction with tetrazine-labeled fixed breast cancer cells to achieve cell labeling. Sulfo-Cy5-TCO is used for fluorescent labeling and imaging of biological samples such as proteins and cells.

Biological Activity
Sulfo-Cy5-TCO undergoes an iEDDA reaction with tetrazine-modified BSA in ammonium acetate buffer at 25 °C to form a fluorescent conjugate, with a yield of approximately 50%[1].
Sulfo-Cy5-TCO can specifically label immobilized MCF7 breast cancer cells with tetrazine tags via the iEDDA reaction, generating detectable red fluorescence, and no non-specific binding is observed under control conditions[1].
