Description

TP4 (Nile tilapia piscidin) is an orally active amphipathic α-helical cationic antimicrobial peptide. TP4 (Nile tilapia piscidin) exerts broad-spectrum bactericidal activity against Gram-positive and Gram-negative bacteria by disrupting cell membrane structures, and also exhibits certain hemolytic activity. In Vitro


TP4 (Nile tilapia piscidin) (10-30 μg/mL; 24 h) inhibits the proliferation of tilapia ovary (TO2) cells in vitro in a dose-dependent manner, and reduces the cell viability to approximately 35% at a concentration of 28 μg/mL[1].
TP4 (Nile tilapia piscidin) (0.3125-40 μg/mL; 48 h) promotes the proliferation of Hs-68 human fibroblasts at concentrations of 2.5, 5, and 10 μg/mL, and does not reduce cell viability at these concentrations[2].
TP4 (Nile tilapia piscidin) (6.25 μg/mL; 48 h) significantly upregulates the gene expression of type I collagen, type III collagen and KGF in the human fibroblast cell line Hs-68[2].
TP4 (Nile tilapia piscidin) (0.3125-40 μg/mL; 48 h) promotes the proliferation of human keratinocyte HaCaT cells at concentrations as low as 10 μg/mL, and does not reduce cell viability within this concentration range[2].
TP4 (Nile tilapia piscidin) (6.25 μg/mL; 48 h) significantly upregulates the expression of keratin 10 gene in human keratinocyte HaCaT cells, but has no effect on the expression of keratin 17[2].

TP4 (0.005 mg/g; i.p.; single injection) provides time-dependent survival benefit in MRSA-septic mice, with 100% survival when administered 10 minutes post-infection, and effectively reduces bacterial burden in all tested organs[2].
TP4 (50-100 mg/kg diet; p.o.; twice daily; 20 days) enhances innate immune responses, antioxidant activity, modulates cytokine gene expression, reduces tissue microbial load (in posterior kidney and intestine), and improves disease resistance against S. iniae in Nile tilapia, with the 100 mg/kg dose conferring the greatest reduction in cumulative mortality (to 30%)[3].
