1-Pyrenebutyric acid is a pyrene-based fluorescent probe compound. The fluorescence lifetime of 1-Pyrenebutyric acid is quenched by molecular oxygen and intracellular ROS, thus it serves as a fluorescent probe for detecting ROS and oxygen concentration through fluorescence lifetime measurements. 1-Pyrenebutyric acid undergoes electrostatic interactions with arginine-rich peptides, increasing their net hydrophobicity, promoting their direct translocation across lipid bilayers, and enhancing cytosolic delivery. 1-Pyrenebutyric acid is used for intracellular reactive oxygen species detection and research related to arginine-rich peptide-mediated cytosolic delivery.

Biological Activity

In Vitro
1-Pyrenebutyric acid (pyrenebutyrate) (50 μM; 2 min) enables rapid, non-membrane-disrupting direct cytosolic and nuclear delivery of 10 μM R8-Alexa peptide in HeLa cells, efficiently delivers 10 μM R8-EGFP protein to the cytosol, and accelerates the internalization of 5 μM R8-PAD peptide, thereby triggering significant mitochondrial depolarization[1].
1-Pyrenebutyric acid (50 μM; 10 min) enhances the cell-killing effect of 5 μM R8-PAD in HeLa cells, reducing cell viability to approximately 20%[1].
1-Pyrenebutyric acid (50 μM; 2 min) rapidly distributes to the perinuclear membrane compartment of HeLa cells and is immediately removed by PBS washing[1].
1-Pyrenebutyric acid (PBA) (0.6 μM; 20 min) exhibits oxygen- and reactive oxygen species-dependent fluorescence lifetime quenching in CCRF-CEM human lymphoblasts, and its fluorescence lifetime signal is independent of the intracellular probe accumulation level[2].
1-Pyrenebutyric acid (0.6 μM; 20 min) detects an 80% increase in reactive oxygen species in CCRF-CEM cells after treatment with Doxorubicin (HY-15142A) (1.7 μM; 3 days), and also detects a 67% decrease in intracellular reactive oxygen species after treatment with cysteamine (1 mM; 3 days)[2].
1-Pyrenebutyric acid (0.6 μM; 20 min) reflects the time-dependent increase in intracellular reactive oxygen species in CCRF-CEM cells following stimulation with exogenous H2O2 (980 μM)[2].
