20S Proteasome-IN-6
20S Proteasome-IN-6 is a species-selective inhibitor of the Mycobacterium tuberculosis 20S proteasome. 20S Proteasome-IN-6 penetrates the Mycobacterium tuberculosis bacilli, sensitizes the bacteria to nitrosative stress, and reduces the bacterial burden of live Mycobacterium tuberculosis within infected macrophages. 20S Proteasome-IN-6 can be used for tuberculosis research.

In Vitro
20S Proteasome-IN-6 (inhibitor 2) is a potent inhibitor of the Mtb 20S proteasome, with a kinact/Ki of 1781 M-1 s-1; it inhibits the β5 subunit of the human 20S proteasome with a kinact/Ki of 17.24 M-1 s-1, exhibiting a species selectivity index of 103-fold[1].
20S Proteasome-IN-6 (0.14‑33.33 μM) penetrates live Mycobacterium tuberculosis and inhibits proteasome activity, thereby effectively stabilizing Pup-Gfp fluorescence[1].
20S Proteasome-IN-6 (33.3 μg/mL; co-incubated with DETA-NO) sensitizes wild-type Mycobacterium tuberculosis H37Rv to nitric oxide-induced nitrosative stress, thereby exerting bacteriostatic-bactericidal effects in the presence of a nitric oxide donor[1].
