Ac-Ala-Ala-Ser (PO3H2)-Pro-Arg-NH-Np (0.3-0.7 mM; time course of cis/trans isomerization) serves as a high-efficiency substrate for purified human Pin1, with a kcat/Km value of 9300 mM-1 s-1; it also acts as a high-efficiency substrate for purified yeast Ess1, with a kcat/Km value of 12000 mM-1 s-1[1].
Ac-Ala-Ala-Ser (PO3H2)-Pro-Arg-NH-Np serves as the optimal-length substrate for purified Pin1 enzyme, while its phosphorylated D-serine analog loses substrate activity[2].
