Ac-Lys (Ac)-D-Ala-D-Ala-OH is a synthetic dipeptide analog that mimics the terminal structure of bacterial peptidoglycan chains. Ac-Lys (Ac)-D-Ala-D-Ala-OH serves as a molecular decoy to investigate the mechanism of action of Vancomycin-class antibiotics, and also acts as an affinity chromatography ligand for purifying proteins that bind to Vancomycin. Ac-Lys (Ac)-D-Ala-D-Ala-OH reduces the activity of Vancomycin against susceptible enterococci. Ac-Lys (Ac)-D-Ala-D-Ala-OH is applicable to the research of Vancomycin-resistant enterococcal infections[1].

In Vitro

Ac-Lys (Ac)-D-Ala-D-Ala-OH (Fig3, A) (20:1, 5:1 peptide:Vancomycin molar ratio; pre-incubation for 1 h, bacterial incubation for 24 h) completely antagonizes the activity of 8 μg/mL and 32 μg/mL Vancomycin (HY-B0671) against Enterococcus faecalis TX2016 at a peptide-to-Vancomycin molar ratio of 20:1, but shows no antagonistic activity at a molar ratio of 5:1[1].
Ac-Lys (Ac)-D-Ala-D-Ala-OH (8 μg/mL vancomycin; 1 hour pre-incubation, 24 hour bacterial incubation) antagonizes the activity of 8 μg/mL vancomycin against Enterococcus faecium TX2016, with an IC50 of 137 μM[1].
