Mechanistically, NEK11 is activated downstream of Chk1 following DNA damage, leading to phosphorylation and degradation of Cdc25A, which enforces the G2/M checkpoint[1][2]. The kinase exists as multiple isoforms, including Nek11L, Nek11S, Nek11C, and Nek11D, which exhibit distinct subcellular localization and differential impact on checkpoint control, with Nek11S showing the strongest effect[1][2]. NEK11 activity is heightened in response to genotoxic stresses such as ionizing radiation and chemotherapeutic agents, promoting cell survival by coordinating DNA repair processes[1][2][3]. In disease contexts, NEK11 overexpression is observed in colorectal and breast cancers, where its modulation influences cell proliferation and apoptosis, making it a potential therapeutic target[1][4].
