Background
Function:Plays a critical role in induction and maintenance of immune tolerance to self (PubMed:11015443, PubMed:28813410, PubMed:28813417, PubMed:31399419). As a ligand for the inhibitory receptor PDCD1/PD-1, modulates the activation threshold of T-cells and limits T-cell effector response (PubMed:11015443, PubMed:28813410, PubMed:28813417, PubMed:36727298). Through a yet unknown activating receptor, may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10) (PubMed:10581077). Can also act as a transcription coactivator: in response to hypoxia, translocates into the nucleus via its interaction with phosphorylated STAT3 and promotes transcription of GSDMC, leading to pyroptosis (PubMed:32929201); The PDCD1-mediated inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity and escape destruction by the immune system, thereby facilitating tumor survival (PubMed:28813410, PubMed:28813417). The interaction with PDCD1/PD-1 inhibits cytotoxic T lymphocytes (CTLs) effector function (By similarity). The blockage of the PDCD1-mediated pathway results in the reversal of the exhausted T-cell phenotype and the normalization of the anti-tumor response, providing a rationale for cancer immunotherapy (By similarity)

Expression:
Tissue_specificity:It is highly expressed in the heart, skeletal muscle, placenta, and lungs. It is weakly expressed in the thymus, spleen, kidneys, and liver. It is expressed on activated T cells and B cells, dendritic cells, keratinocytes, and monocytes; it is widely expressed, with the highest expression in the lungs, liver, pituitary gland, and various peripheral blood cells (including neutrophils and certain subtypes of lymphocytes and myeloid cells) .

Induction:Up-regulated on T- and B-cells, dendritic cells, keratinocytes and monocytes after LPS and IFNG activation. Up-regulated in B-cells activated by surface Ig cross-linking
Isoforms & Post-Translational Modification:Q9NZQ7 has 4 isomers: Q9NZQ7-1: 33275 Da (predicted); Q9NZQ7-2: 20246 Da (predicted); Q9NZQ7-3: 20454 Da (predicted); Q9NZQ7-4: 28050 Da (predicted).
Ubiquitinated; STUB1 likely mediates polyubiquitination of PD-L1/CD274 triggering its degradation (PubMed:28813410). Ubiquitinated by MARCHF8; leading to degradation (PubMed:34183449). Deubiquitinated by USP22; leading to stabilization (PubMed:31399419)
Subunit:Interacts with PDCD1 (PubMed:11015443, PubMed:18287011, PubMed:26602187). Interacts (via transmembrane domain) with CMTM4 and CMTM6 (PubMed:28813410, PubMed:28813417). Interacts with (phosphorylated) STAT3; promoting nuclear translocation (PubMed:32929201). Interacts with CD80 (PubMed:36727298)

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
