U 66985 is a platelet-activating factor (PAF) receptor antagonist. U 66985 binds to the PAF receptor site, thereby inhibiting PAF-induced platelet aggregation. U 66985 can be used in studies on the PAF signaling pathway and the functions of lipid inflammatory mediators.

Activité biologique
U 66985 (radioimmunoassay; incubation at room temperature overnight) inhibits the binding of [125I]PAF to anti-PAF antibody from rabbit A, with a 50% inhibitory amount of 19 pmol/tube[1]
U 66985 (1 μM; HAG 10 μg/mL; pre-perfused for 5 min prior to HAG stimulation; isolated perfused liver of Sprague-Dawley rats) does not reduce the HAG-induced glycogenolysis response; the maximum increase in glucose release upon U 66985 treatment is 81 μmol/h/g, compared with 80 μmol/h/g in the parallel control group[2].
U 66985 (1 μM; isolated perfused Sprague-Dawley rat liver; pretreated starting at 35 min of perfusion, followed by perfusion with 1 nM [3H]18:0-sn-3-AGEPC during 40-42 min) does not significantly alter the silver grain distribution of [3H]AGEPC in liver tissues; in this experiment, U 66985 is present at a 1000-fold excess relative to the radioligand[3].
