DMTr-2'-O-C22-rG (ibu)-3'-CE-Phosphoramidite supports target gene silencing in skeletal muscle, cardiac muscle and adipose tissue by enhancing the lipid solubility and tissue uptake efficiency of dsRNA. DMTr-2'-O-C22-rG (ibu)-3'-CE-Phosphoramidite (Compound 106) is synthesized via phosphoramidite coupling reaction with 2-cyanoethyl N,N-diisopropylchlorophosphoramidite, monitored by TLC (Rf = 0.6 in 50% ethyl acetate/hexane), purified by silica gel column chromatography to obtain a white solid with 73% yield (1.76 g), and characterized by 1H NMR (600 MHz, CD3CN), 31P NMR (243 MHz, CD3CN) and LC-MS (ESI) with observed [M+H]⁺ = 1164.8[1].
