DMTr-2'-O-C22-rC (Ac)-3'-CE-Phosphoramidite is an RNA phosphoramidite monomer. DMTr-2'-O-C22-rC (Ac)-3'-CE-Phosphoramidite enables site-specific introduction of 2'-O-C22 lipophilic modification at cytidine positions during oligonucleotide synthesis, which is used to construct double-stranded RNA (dsRNA) molecules with extrahepatic delivery capability. In Vitro

DMTr-2'-O-C22-rC (Ac)-3'-CE-Phosphoramidite (Compound 114) is synthesized via a phosphoramidite coupling reaction with 2-cyanoethyl N,N-diisopropylchlorophosphoramidite. The reaction progress is verified through work-up and purification procedures. After quenching with saturated sodium bicarbonate, extraction with ethyl acetate, washing with saturated sodium chloride, and drying over anhydrous sodium sulfate, the crude product is purified by silica gel column chromatography to yield a white foamy solid. The product is characterized by 1H NMR (600 MHz, DMSO) and 31P NMR (243 MHz, DMSO), with 31P NMR chemical shifts measured at δ 149.23 and 147.94[1].
