FA-Leu-Gly-Pro-Ala-OH is a furylacryloyl-terminal tetrapeptide that serves as a substrate for bacterial collagenase and spirochete metalloendopeptidase. FA-Leu-Gly-Pro-Ala-OH is specifically hydrolyzed by spirochete collagenase only at the Leu-Gly bond. FA-Leu-Gly-Pro-Ala-OH can be used to determine the equilibrium constant of peptide bond hydrolysis, and also to detect collagenase-mediated cleavage reactions via turbidimetry based on absorbance reduction.

Biological Activity
FA-Leu-Gly-Pro-Ala-OH (1 mM Ca2+; no added Ca2+) is hydrolyzed at the Leu-Gly bond by the partially purified metalloendopeptidase from unspeciated treponeme strain US with near-identical activity in the presence of 1 mM Ca2+ or without added Ca2+[1].
FA-Leu-Gly-Pro-Ala-OH (5 mM; 15 min) was not cleaved by crude nematocyst extract from Velella velella, which did not exhibit detectable collagenase activity against the substrate under the tested conditions[2].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
