Flow cytometric analysis of 1X106 HUVEC cells labeling Merinetug (HY-P991072, red).Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4°C. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776)was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4°C. Human IgG1 kappa, Isotype Control (HY-P99001 blue) was used as the isotype control,cells without incubation with primary antibody were used as thunlabeled control (black).

Amyloid: Congo Red Amyloid Staining
Congo red amyloid staining is a histochemical method used to detect extracellular amyloid deposits in tissue sections based on the affinity of Congo red dye for β-pleated sheet-rich protein aggregates. When bound to amyloid, Congo red produces characteristic apple-green birefringence under polarized light microscopy, which is widely regarded as a diagnostic feature of amyloid deposition in histopathology. The diagnostic principle relies on the combination of dye binding (congophilia) and optical anisotropy under polarized illumination, which distinguishes amyloid from most non-amyloid eosinophilic extracellular deposits in routine histological evaluation.
