Ac-PKM-AMC is a fluorescent tripeptide substrate for ClpP1P2 peptidase (excitation and emission: 380/460 nm). Ac-PKM-AMC is used to measure Mtb ClpP1P2 peptidase activity^[1]^[2]^[3].

In Vitro

Ac-PKM-AMC (100 μM) serves as the fluorogenic substrate for measuring Mtb ClpP1P2 peptidase activity, with cleavage monitored by fluorescence at 380/460 nm^[1].
Ac-PKM-AMC (10 μM; at least 1 h) is the substrate used in the ClpP1P2 peptidase degradation assay to determine IC₅₀ values of inhibitors^[1].
Ac-PKM-AMC hydrolysis by Mtb ClpP1P2 is dramatically stimulated by Bz-LL or Bz-Nva-Ile in the presence of ClpX or ClpC1, with half-maximal activation requiring an order of magnitude lower activator concentration when either ATPase is present^[2].
