1. Metabolic Enzyme/Protease
  2. E1/E2/E3 Enzyme Heme Oxygenase (HO)
  3. N,N-dimethyldithiocarbamate potassium

N,N-dimethyldithiocarbamate potassium is a ubiquitin-activating enzyme E1 inhibitor. N,N-dimethyldithiocarbamate potassium inhibits the activation of E1 and reduces the level of activated ubiquitinated E1 (ub-E1). N,N-dimethyldithiocarbamate potassium increases intracellular copper levels, enhances oxidative stress, elevates protein carbonyl levels, and upregulates the expression of HO-1. N,N-dimethyldithiocarbamate potassium is applicable to research related to Parkinson's disease.

Description

N,N-dimethyldithiocarbamate potassium is a ubiquitin-activating enzyme E1 inhibitor. N,N-dimethyldithiocarbamate potassium inhibits the activation of E1 and reduces the level of activated ubiquitinated E1 (ub-E1). N,N-dimethyldithiocarbamate potassium increases intracellular copper levels, enhances oxidative stress, elevates protein carbonyl levels, and upregulates the expression of HO-1[1]. N,N-dimethyldithiocarbamate potassium is applicable to research related to Parkinson's disease[1].

In Vitro

N,N-dimethyldithiocarbamate (potassium) (20 μM; 6 h) inhibits ubiquitin-activating enzyme E1 activation in HEK293 cells, reducing the ub-E1/E1 ratio to 0.25 relative to control, with no detectable covalent E1 adducts[1].
N,N-dimethyldithiocarbamate (20 μM; 6 h) potassium significantly increases heme oxygenase-1 protein expression, a marker of oxidative stress, in HEK293 cells[1].
N,N-dimethyldithiocarbamate (20 μM; 6 h) potassium significantly elevates intracellular copper levels in HEK293 cells[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Western Blot Analysis[1]

Cell Line: HEK293 cells
Concentration: 20 μM
Incubation Time: 6 h
Result: Significantly decreased the ub-E1/E1 ratio to 0.25 relative to control (p < 0.01, n = 5).
Detected no dithiocarbamate-derived E1 adducts (S-methylaminocarbonyl or glutathiotinylation) in E1-enriched cell extracts via LC/MS/MS. Significantly increased protein carbonyl levels relative to control (p < 0.05, n = 5). Significantly increased HO-1 protein expression relative to control (p < 0.01, n = 5).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
