Peanut Agglutinin (PNA) Fluorescein is a fluorescently labeled lectin that specifically binds to glycosylated cells and Haemonchus contortus. Peanut Agglutinin Fluorescein specifically recognizes and binds to the D-Gal-β(1→3)-D-GalNAc disaccharide sequence and terminal D-galactosyl residues on the cell surface; its binding can be inhibited by D-galactose but is not affected by glucose. Peanut Agglutinin Fluorescein specifically labels cone photoreceptor segments and can also be used to distinguish immature thymocytes from lymphocyte subsets, serving as an effective tool for studying cellular glycosylation status and isolating specific cell populations.

Biological Activity
Fluorescein-conjugated Peanut Agglutinin (FITC-PNA) (100-200 μg/mL; 15-30 min) binds specifically to cone photoreceptor inner and outer segments in human, monkey, rabbit, mouse, chicken, and fish retinas, with additional species-specific binding to cone-associated synaptic structures, and this binding is galactose-dependent[1].
Fluorescein-conjugated Peanut Agglutinin (FITC-PNA) (200 μg/mL; 30 min) binds specifically to cone outer segment tips and cone inner/outer segment plasma membranes in whole-mount rabbit retinas, with no binding to rod photoreceptors[1].
Peanut Agglutinin Fluorescein (0.5 mg/mL, 5 μg; 30 min) stains 80%-90% of isolated thymocytes and 2.7%-15.8% of isolated peripheral lymphocytes; the proportion of labeled cells is higher among dense adult CBA/J thymocytes than among low-density thymocytes. Furthermore, this reagent detects cells at the early stages of lymphopoiesis in isolated fetal cells, serving as a marker for immature or early T-cell subpopulations[2].

Peanut Agglutinin Fluorescein (5 mg/mL; 1 h) maintains specific binding to Haemonchus contortus eggs fixed with 0.5-5% formalin and stored at 4°C for no more than 4 weeks, while a decrease in staining consistency is observed after 5 weeks of storage[3].
