Description

The acid prostatic phosphatase (ACPP) protein, is an acid tyrosine phosphatase that skillfully dephosphorylates a variety of substrates. ACPP-VLP possesses lipid phosphatase activity and inactivates lysophosphatidic acid. ACPP/Prostatic Acid Phosphatase Protein, Mouse (HEK293, His) is the recombinant mouse-derived ACPP/Prostatic Acid Phosphatase protein, expressed by HEK293 , with C-His labeled tag.

Background

Prostatic Acid Phosphatase (ACPP) protein serves as a versatile non-specific tyrosine phosphatase, exhibiting its catalytic activity under acidic conditions (pH 4-6) on a broad range of substrates, including alkyl, aryl, and acyl orthophosphate monoesters, as well as phosphorylated proteins. Notably, ACPP also displays lipid phosphatase activity, contributing to the inactivation of lysophosphatidic acid in seminal plasma. Beyond its tyrosine phosphatase functions, ACPP demonstrates ecto-5'-nucleotidase activity in dorsal root ganglion neurons, leading to the generation of adenosine from AMP. This extracellular adenosine, in turn, contributes to a reduction in chronic pain by activating A1R in nociceptive neurons. The multifaceted enzymatic activities of ACPP highlight its potential significance in various cellular processes and its therapeutic implications, particularly in pain management.

Measured by its ability to cleave 2mM substrate p-Nitrophenyl phosphate (pNPP). which can be measured by absorbance at 410 nm that incubate at room temperature for 5 minutes. The specific activity is 104600.068 pmol/min/µg, as measured under the described conditions.

Assay Procedure

Materials
Assay buffer: 50 mM NaOAc, pH 4.5
Stop solution: 0.2 M NaOH
Substrate: 2 mM pNPP (HY-116022A)
Standard: 4-Nitrophenol

Procedure

1. Dilute 4-Nitrophenol in assay buffer to the following concentrations: 0, 0.03125, 0.0625, 0.125, 0.25, 0.5, 1 mM.
2. Add 100 μL of each standard concentration to a well.
3. Incubate at room temperature for 5 minutes.
4. Add 100 μL of 0.2 M NaOH to stop the reaction and develop the color.
5. Immediately read the absorbance at 410 nm in endpoint mode.
6. Plot the standard concentration on the y-axis and the measured OD value on the x-axis to generate a standard curve and obtain the standard curve equation.
7. Dilute Human ACPP to 0.1 μg/mL.
8. Dilute the substrate to 2 mM in assay buffer.
9. In a 96-well plate, add 50 μL of 0.1 μg/mL Human ACPP and 50 μL of 2 mM pNPP. Include a blank control with 50 μL assay buffer and 50 μL of 2 mM pNPP. Perform the experiment in triplicate.
10. Incubate the reaction at room temperature for 5 minutes.
11. Add 100 μL of 0.2 M NaOH to stop the reaction and develop the color.
12. Measure absorbance at 410 nm.
