Description

AURA is a mitotic serine/threonine kinase that complexly controls cell cycle progression and various mitotic events. AURA associates with centrosomes and spindle microtubules during mitosis and plays a critical role in spindle establishment, centrosome duplication, chromosome alignment, spindle assembly checkpoint activation, and cytokinesis. AURKA Protein, Human (Active, sf9, His-GST) is the recombinant human-derived AURKA, expressed by Sf9 insect cells, with N-6*His and N-GST labeled tag.

Background

AURA, a mitotic serine/threonine kinase, intricately regulates cell cycle progression and contributes significantly to various mitotic events. During mitosis, AURA associates with the centrosome and spindle microtubules, playing a critical role in spindle establishment, centrosome duplication, separation, chromosomal alignment, spindle assembly checkpoint activation, and cytokinesis. Essential for proper spindle positioning and the localization of NUMA1 and DCTN1 to the cell cortex during metaphase, AURA is also required for the initial activation of CDK1 at centrosomes. Functionally versatile, AURA phosphorylates a myriad of target proteins involved in diverse cellular processes, including microtubule dynamics, axon formation, and neurite extension. Moreover, AURA acts as a pivotal regulator of the p53/TP53 pathway, phosphorylating and destabilizing p53/TP53, and influences cilia dynamics by inhibiting cilia outgrowth and promoting cilia disassembly. Additionally, AURA plays a role in modulating the protein levels of the anti-apoptosis factor BIRC5 through the phosphorylation of the transcription factor FOXP1, highlighting its multifaceted impact on cellular processes.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
