Background

In the context of retinal vascularization, Frizzled-4 plays a pivotal role by acting as a receptor for both Wnt proteins and norrin (NDP), facilitating beta-catenin accumulation and stimulation of LEF/TCF-mediated transcriptional programs. Although a secondary signaling pathway, featuring PKC and calcium fluxes, has been observed in some family members, its integration with the canonical pathway, particularly in the context of Wnt-mediated inactivation of GSK-3 kinase, remains uncertain. Frizzled-4's involvement in tissue morphogenesis and intercellular transmission of polarity information is suggested, with interactions observed with MAGI3, NDP, TSKU, and glypican GPC3, highlighting its intricate role in cellular processes and signaling cascades.
Verified Bioactivity
Measured by its binding ability in a functional ELISA. Immobilized Human Frizzled-4, at 0.1 μg/mL (100 μL/well) can bind Wnt-5a. The ED50 for this effect is 26.59 ng/mL.
