Nectin-2/CD112 protein has dual roles in T-cell signaling: as a costimulator with CD226, promoting proliferation and cytokine production, and as a coinhibitor with PVRIG, inhibiting proliferation. It also acts as a cell adhesion protein and a receptor for certain viruses. Furthermore, in the context of microbial infection, it acts as a receptor for herpes simplex virus 1 (HHV-1) mutant Rid1, herpes simplex virus 1 (HHV-2), and pseudorabies virus (PRV).
