Background

PARP2 is a poly-ADP-ribosyltransferase that mediates protein poly-ADP-ribosylation and plays a key role in DNA repair. It catalyzes glutamate, aspartate, or serine ADP-ribosylation, transferring the ADP-D-ribosyl group from NAD(+) to target protein carboxyl groups and forming polymers with an average length of 20-30 units. Serine ADP-ribosylation is the primary form in DNA damage response. Without HPF1, PARP2 mediates glutamate and aspartate ADP-ribosylation, while HPF1 binding confers serine specificity. PARP2 recognizes DNA breaks in chromatin, recruits HPF1, and promotes serine ADP-ribosylation of histones, decompacting chromatin and recruiting repair factors. HPF1 restricts PARP2's polymerase activity to limit poly-ADP-ribose chain length. PARP2 also specifically mediates branched poly-ADP-ribosylation, recognized by factors like APLF. Additionally, PARP2 can ADP-ribosylate DNA, preferentially acting on 5'-terminal phosphates at DNA strand breaks in nicked duplexes.
