2,6-Dichloro-4-nitroaniline is an orally active Herbicide, Fungicide and uncoupler. 2,6-Dichloro-4-nitroaniline uncouples oxidative phosphorylation and inhibits electron transport. 2,6-Dichloro-4-nitroaniline induces biphenyl hydroxylase activity in rat liver. 2,6-Dichloro-4-nitroaniline increases the relative liver weight of rats via hepatomegaly without altering their body weight[1].

In Vitro

2,6-Dichloro-4-nitroaniline (1×10-5-1×10-4 M) uncouples oxidative phosphorylation and inhibits electron transport in isolated liver mitochondria[1].

2,6-Dichloro-4-nitroaniline (500-1000 mg/kg; p.o.; single dose; administered for 4 consecutive days; up to 12 consecutive weeks) induces hepatomegaly and biphenyl hydroxylase activity in female Sprague-Dawley rats[1].

Animal Model: Sprague-Dawley (female, 100-150 g)[1]
Dosage: 500-1000 mg/kg; 1000 mg/kg (single dose); 1000 mg/kg (daily for 4 days); 500 mg/kg (daily for up to 12 weeks)
Administration: p.o.; single dose; daily (4 consecutive days); daily (up to 12 weeks)
Result: Increased hepatic mitochondrial respiratory rate by 18% (16 hours post single 1000 mg/kg dose, succinate as substrate).
Did not alter ADP/O ratio at 1, 16, or 24 hours post single 1000 mg/kg dose.
Increased hepatic 4-OH biphenyl hydroxylase activity to 2.545 μmol/hr/g fresh liver and 2-OH biphenyl hydroxylase activity to 0.513 μmol/hr/g fresh liver (24 hours post single 1000 mg/kg dose).
Increased hepatic mitochondrial respiratory rate to 54.15, 50.94, and 44.53 nmol O2/min/mg protein (1000 mg/kg daily for 2, 3, 4 days respectively, succinate as substrate).
Did not alter ADP/O ratio (1000 mg/kg daily for 2, 3, 4 days).
Increased relative liver weight to 7.10, 7.43, and 6.10 (1000 mg/kg daily for 2, 3, 4 days respectively).
Increased hepatic 4-OH biphenyl hydroxylase activity to 3.705, 5.205, and 4.275 μmol/hr/g fresh liver and 2-OH biphenyl hydroxylase activity to 3.715, 5.181, and 4.385 μmol/hr/g fresh liver (1000 mg/kg daily for 2, 3, 4 days respectively).
Increased relative liver weight to 6.7, 8.1, 6.1, and 5.6 (500 mg/kg daily for 2, 4, 8, 12 weeks respectively).
Increased hepatic 4-OH biphenyl hydroxylase activity to 1.710, 2.27, 1.16, and 3.46 μmol/hr/g fresh liver and 2-OH biphenyl hydroxylase activity to 2.378, 1.48, 1.05, and 4.45 μmol/hr/g fresh liver (500 mg/kg daily for 2, 4, 8, 12 weeks respectively).
Elevated respiratory activity in 33,000 g·min heavy mitochondrial pellet throughout 3 months of 500 mg/kg daily treatment.
Detected ~1×10-5 M DCNA in whole liver homogenate and crude nuclear pellet (500 mg/kg daily for 4 days).
Detected 37.2 μg/mL DCNA in urine (single 1000 mg/kg dose) and 30.6 μg/mL DCNA in urine (500 mg/kg daily for 30 days).
