MuRF1-IN-1 is an orally active MuRF1 inhibitor. MuRF1-IN-1 can inhibit the interaction between MuRF1 and titin as well as E3 ligase activity. MuRF1-IN-1 can alleviate skeletal muscle atrophy and dysfunction in cardiac cachexia.

Result:Prevented atrophy in tibialis anterior (TA) muscle, with preserved wet weight and fibre cross-sectional area (CSA) compared to monocrotaline-treated controls. Wet weights of extensor digitorum longus (EDL) and soleus muscles were normalized to sham levels.
Restored diaphragm muscle shortening velocity and power to sham levels, reversing the ~20% impairment induced by cardiac cachexia.
Downregulated MuRF1 protein expression, normalized pro-apoptotic protein BAX and translation initiation factor eIF2B-δ, and attenuated actin ubiquitination and proteasome activity.
