RMC-3943 is an allosteric SHP2 inhibitor (inhibition of full-length SHP2 in biochemical assay, IC₅₀ = 2.19 nM)^[1].

In Vitro

RMC-3943 inhibits pERK in PC9 cells, with an IC₅₀ of 35.5 nM^[1].
RMC-3943 (0-10 μM) produces a concentration-dependent inhibition of proliferation of SK-MEL-113 cells^[1].
