Clobetasol

Clobetasol is a glucocorticoid and Smoothened agonist that can cross the blood-brain barrier. Clobetasol induces Smo-dependent activation of the Shh signaling pathway and nuclear translocation of Gli1. Clobetasol promotes remyelination and reduces myelin loss in lesions of neuromyelitis optica (NMO). Clobetasol may cause adrenal suppression, cushingoid appearance, and local adverse reactions including dysgeusia, xerostomia, and gastroesophageal burning. Clobetasol can be used in research related to neurodegenerative diseases, oral lichen planus, mucous membrane pemphigoid, and neuromyelitis optica.

Clobetasol (0.1-25 µM) enhances the metabolic turnover of neural stem cells (NSCs) derived from the subventricular zone (SVZ) of mice via a Smo-dependent signaling pathway[1].
Clobetasol (5 µM) doubles the cell growth rate of neural stem cells (NSCs) derived from the subventricular zone (SVZ) of mice via the Smo-dependent signaling pathway[1].
Clobetasol increases Gli1 mRNA levels in mouse SVZ-derived neural stem cells (NSCs) via the Smo-dependent signaling pathway[1].
Clobetasol (0.3-10 μM; 6 days) promotes the maturation of primary mouse oligodendrocyte precursor cells, and approximately 15% of the cells become MBP-positive after incubation at 5 μM for 6 days[3].
Clobetasol (6 days) promotes remyelination and increases the number of mature oligodendrocytes in cultured mouse cerebellar slices with demyelination induced by AQP4-IgG and complement after 6 days of treatment[3].

Did not significantly alter initial astrocyte damage (AQP4 loss), inflammation (Iba1 and CD45 immunofluorescence), or complement activation (C5b-9 deposition) compared to vehicle.
Produced significantly greater remyelination than vehicle when administered starting on day 4 (post-demyelination).
