Pifexole
Pifexole is a biologically active 3,5-disubstituted-1,2,4-oxadiazole compound. Pifexole exhibits antispasmodic and muscle relaxant activities, with no effect on monosynaptic knee-jerk reflex. Pifexole can be used in studies related to muscle relaxation and spasm.

Activité biologique
Pifexole (15.4-200 mg/kg; p.o.) inhibits electroshock-induced convulsions in rats and mice with ED50 values of 17.5 mg/kg and 15.4 mg/kg, respectively, and inhibits strychnine-induced convulsions in both species with an ED50 of 96 mg/kg, while antagonizing nicotine toxicity in mice with an ED50 of 30 mg/kg. Pifexole also reduces muscle strength in mice by 25% and 35% at 100 mg/kg and 200 mg/kg, respectively, inhibits spontaneous activity in mice at 100 mg/kg, and reduces morphine-induced excitation and hyperactivity at 200 mg/kg[2]

Animal Model:Polysynaptic crossed extensor and monosynaptic patellar reflex assays in chloralose-urethane-anesthetized cats[2]
-
Dosage:5, 10, 20 mg/kg
-
Administration:i.v.; single administration; monitored for approximately 20-25 min
-
Result:Reduced the polysynaptic crossed extensor reflex by 81.6% and 93.1% at 5 and 10 mg/kg, respectively.
Did not affect the monosynaptic patellar reflex at 10-20 mg/kg.

Animal Model:Nicotine toxicity[2]
-
Dosage:30, 100, 200 mg/kg
-
Administration:p.o. for the nicotine toxicity assay
-
Result:Antagonized nicotine toxicity with an ED50 of 30 mg/kg.
Reduced muscle strength by 25% and 35% at 100 and 200 mg/kg, respectively.
Suppressed spontaneous locomotor activity at 100 mg/kg and reduced morphine-induced excitation and hypermotility at 200 mg/kg.
