Florfenicol amine hydrochloride is the major metabolite of Florfenicol (HY-B1374) and serves as a marker residue in studies of the in vivo disposition and residue depletion of Florfenicol. Florfenicol amine hydrochloride is eliminated more slowly than the parent compound and persists in plasma and tissues. Florfenicol amine hydrochloride is used in veterinary research for studies on the metabolism, pharmacokinetics, and residue depletion of Florfenicol.

In Vivo
Florfenicol amine hydrochloride is formed and detected in plasma following a single 20 mg/kg dose of Florfenicol administered intravenously or orally via nasogastric tube to 12 healthy male New Zealand White rabbits; after maternal intravenous administration, the Cmax of Florfenicol amine hydrochloride is 5.06 μg/mL, the tmax is 0.88 h, the t1/2 is 1.84 h, the AUC is 20.59 μg·h/mL, and the MRT is 3.15 h; after maternal oral administration, the Cmax is 3.38 μg/mL, the tmax is 2.10 h, the t1/2 is 2.35 h, the AUC is 17.82 μg·h/mL, and the MRT is 4.28 h[2].
In a single-dose study of Atlantic salmon (Salmo salar) in seawater at approximately 10°C, Florfenicol amine hydrochloride is detected in plasma following intravenous or oral administration of florfenicol at 10 mg/kg; from 48 h post-dose until the end of the study, its plasma concentration is higher than that of florfenicol, and regression analysis estimates its plasma elimination half-life to be 23 h[3].
Florfenicol amine hydrochloride is formed in Atlantic salmon in a multiple-dose study in which Florfenicol is administered at 10 mg/kg daily with feed for 10 consecutive days, and the plasma elimination half-life after the end of the multiple-dose period is estimated to be 24.2 h[3].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.08 mg/mL (7.33 mM); Clear solution
This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.08 mg/mL (7.33 mM); Clear solution
This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
