ICG-carboxylic acid is near-infrared (NIR) fluorescent probe. ICG is a fluorescent dye used in medical diagnostics. ICG has absorption peaking at 800 nm and can absorb the near IR laser energy and release heat in the dyed tissue (Ex/Em = 785/810 nm).

Biological Activity
Guidelines (The following is our recommended protocol; this protocol serves merely as a guideline and should be modified according to your specific needs)[3]
1. Add 1 mg of ICG-carboxylic acid to 10 mL of water containing 100 mg of the aluminum-rich polysaccharide “SpaTA”; subsequently, mix with 50 mg of EDC/NHS and allow the reaction to proceed catalytically at room temperature for 6 hours.
2. Transfer the solution into a micro-dialysis bag, then purify it using 10 mM PBS.
3. Replace the dialysis buffer every 4 hours, for a total of three changes.
4. Determine the Relative Fluorescence Units (RFU) of the ICG-loaded SpaTA and the ICG standard using fluorescence spectrophotometry, with an excitation wavelength of 805 nm and an emission wavelength of 835 nm.
5. The formula for calculating the ICG Loading Rate (LR) is as follows:
LR% = RFUSample / RFUStandard.
