BMX-IN-4 (Compound 11i) potently inhibits recombinant BMX with an IC₅₀ of 1.78 nM; it exhibits 2.3-fold selectivity over recombinant BTK, and its activity is dependent on a covalent warhead^[1].
BMX-IN-4 achieves selective cellular target binding to BMX but not BTK in HEK293T cells, with an EC₅₀ of 0.098 μM against BMX and a selectivity window of 20.6-fold^[1].
