L-BPA-BPA hydrochloride is an aromatic dipeptide and an analog of L-BPA (HY-W087830). L-BPA-BPA hydrochloride undergoes rapid hydrolytic cleavage in vivo to release the boron neutron capture agent L-BPA. L-BPA-BPA hydrochloride enables boron delivery to tumors; when used in combination with neutron irradiation, it also induces immunological effects including vaccine responses and abscopal tumor suppression. L-BPA-BPA hydrochloride can be used in the research of colon cancer and hypopharyngeal cancer.

Result:Delivered higher tumor boron concentrations compared to 200 mg/kg L-BPA.
Suppressed tumor growth at day 25 with 12-minute neutron irradiation.
Maintained robust tumor suppression with 6-minute neutron irradiation, while 200 mg/kg L-BPA only delayed growth.
Produced durable complete tumor regression in all treated mice through day 43 with 6-minute neutron irradiation, with no viable tumor cells detected.
Induced a vaccine effect: all mice rejected CT26 cell rechallenge in the contralateral hind leg, while naïve mice developed progressive tumors.
Induced marked regression of the irradiated index tumor and a 34% growth delay in the unirradiated contralateral tumor in a bilateral tumor model.
