Laprafylline (S 9795), a xanthine derivative, is a bronchodilator. Laprafylline has potent anti-bronchoconstrictive effects, inhibiting action on mast cell degranulation and phosphodiesterase (PDE) activity (IC50 of 6 μM)[1].

In Vivo

In male Sprague Dawley rats, Laprafylline (S 9795; 2.5-10mg/kg; iv) disappears rapidly from plasma with an elimination t1/2 of 0.8 h, regardless of the dose administered. The volume of distribution is high (about 3 L/kg) but total clearance is also high (about 40 mL/min/kg).
