Background
CTNNA1 is a protein that associates with the cytoplasmic domain of various cadherins. Its binding to cadherins forms a complex linked to the actin filament network, which is crucial for the cell-adhesion properties of cadherins. CTNNA1 can interact with both E-cadherin and N-cadherin. Initially thought to be a stable component of E-cadherin/catenin adhesion complexes and to mediate the connection between cadherins and the actin cytoskeleton at adherens junctions, later studies revealed that cortical actin is more dynamic than E-cadherin/catenin complexes, and CTNNA1 does not bind to F-actin within the complex, suggesting an alternative linkage mechanism between actin and adherens junction components. The homodimeric form of CTNNA1 may regulate actin filament assembly and inhibit actin branching by competing with the Arp2/3 complex for actin filament binding. It is also involved in the regulation of WWTR1/TAZ, YAP1, and TGFB1-dependent SMAD2 and SMAD3 nuclear accumulation (By similarity) and may play a critical role in cell differentiation.
